表皮生长因子(EGFs)

多种疾病与生长因子稳态的破坏相关,表皮生长因子(EGF)家族就是如此,其表达异常与多种疾病的发生有关,如肺腺癌和其他多种癌症类型、慢性疼痛、慢性肾脏疾病和银屑病。EGF生长因子的信号传导由ErbB家族的G蛋白偶联受体介导,该家族包含四个成员(ErbB1至ErbB4)。EGF生长因子与受体的结合由EGF结构域介导,导致受体的同源或异源二聚化,并激活其细胞内C末端结构域的交叉磷酸化。受体激活可触发信号转导至参与分化、增殖、迁移或存活的多条通路,具有广泛的靶点,包括PLCϒ、STAT5、SHIP1、Abl、PTP-2c、STAT1、Syk、Vav2、RasA1、SH3BGRL、Grb2、Src、Crk、Nck、Shc和Cb1。
