胰岛素样生长因子(IGFs)

IGF家族主要由两种低分子量蛋白质(IGF-1和IGF-2)、相应的受体(IGF-1R和IGF-2R)以及特异性结合蛋白组成。1963年,Froesch等人在血清中发现了一种不能被胰岛素抗体完全抑制的活性物质;随后,Rinderknecht和Humbel两位科学家对其进行了纯化,并命名为IGF-1和IGF-2。 胰岛素样生长因子1(IGF1)和胰岛素样生长因子2(IGF2)可以与其受体胰岛素样生长因子1受体(IGF1R)结合,随后通过磷酸化胰岛素受体底物并与磷脂酰肌醇3-激酶(PI3K)和生长因子受体结合蛋白2相关蛋白(Grb2)结合,触发下游级联反应,从而激活两个重要的信号通路:(1)丝裂原活化蛋白激酶(MAPK)通路,主要诱导转录和代谢中的各种反应,调节细胞生长、存活和分化。(2)PI3K通路,参与细胞存活、生长和增殖,以及蛋白质合成。
