白细胞介素(ILs)及其受体

白细胞介素(ILs)是一组细胞因子(分泌蛋白和信号分子),最初被发现由白细胞(leukocytes)表达。免疫系统的功能在很大程度上依赖于白细胞介素,已经描述了多种白细胞介素的罕见缺陷,这些缺陷都表现为自身免疫疾病或免疫缺陷。大多数白细胞介素由辅助性CD4 T淋巴细胞合成,也可通过单核细胞、巨噬细胞和内皮细胞合成。它们促进T和B淋巴细胞以及造血细胞的发育和分化。许多白细胞介素根据结构相似性、受体链使用和功能特性被分类为不同的家族。例如IL-1家族、IL-2家族、IL-6家族、IL-10家族、IL-12家族、IL-17家族等。每个家族都有独特的特征,在免疫调节和炎症中发挥特定的作用。
白细胞介素受体(ILRs)是一类广泛表达于免疫细胞和非免疫细胞表面的跨膜蛋白家族。其核心功能是特异性结合白细胞介素(ILs)并启动细胞内信号转导,从而调节机体免疫应答、细胞增殖与分化以及生理和病理过程。目前已鉴定的38种白细胞介素(IL-1至IL-38)均需与相应的ILRs结合才能发挥作用,两者之间的特异性结合是白细胞介素功能实现的首要步骤。
根据结构特征和信号传导机制,ILRs家族主要分为四类:第一类是IL-17受体家族,包含五个成员(IL-17RA至IL-17RE)。它们多以复合物形式(如IL-17RA/RC复合物)介导促炎和免疫应答信号,保守的胞内"SEFIR"结构域是信号转导的关键基础;第二类是Ig样受体,属于免疫球蛋白超家族(IgSF),包含1-7个Ig样结构域。典型成员包括IL-1受体和IL-18受体,广泛参与炎症启动和免疫调节;第三类是I型受体(又称红细胞生成素受体家族),大多由多个亚基组成。其胞外区含有保守的半胱氨酸残基和WSXWS序列(Trp-Ser-X-Trp-Ser),涵盖IL-2、IL-3、IL-6等受体,调控免疫细胞增殖和造血;第四类是II型受体(又称干扰素受体家族),其胞外区含有四个保守的半胱氨酸残基但无WSXWS序列。它们多以异二聚体形式存在,如IL-10和IL-22受体,常与Janus激酶(JAK)结合传递抗炎或抗病毒信号。
