干扰素(IFNs)

干扰素(IFN)家族包含三类:I型(IFN-α/β/κ/ε/ω)、II型(IFN-γ)和III型(IFN-λ1/2/3)。I型IFN在病毒或核酸感知后迅速产生,通过IFNAR1/2信号激活JAK-STAT1/2通路,诱导数百个干扰素刺激基因(ISG)的表达,建立抗病毒状态。II型IFN由NK细胞和Th1细胞分泌,通过IFNGR1/2增强巨噬细胞激活和抗原提呈。III型IFN通过IFNLR1/IL-10R2在黏膜表面局部发挥作用,同样使用STAT1/2通路。除抗病毒防御外,IFN还调节细胞增殖、凋亡和肿瘤免疫监视;其失调是慢性感染、自身免疫和癌症免疫逃逸的基础,使其成为基于IFN的疗法和免疫检查点策略的核心靶点。
