成纤维细胞生长因子(FGFs)

成纤维细胞生长因子(FGF)家族由一组结构相关的信号蛋白组成,在胚胎发育、细胞增殖、分化、迁移、血管生成、组织修复和代谢调节中发挥关键作用。在人类中,FGF家族包括18个经典成员(FGF1–FGF10和FGF16–FGF23),根据序列同源性和功能特征分为六个亚家族。大多数FGF以旁分泌方式在局部发挥作用,而FGF19亚家族(FGF19、FGF21和FGF23)则以内分泌方式发挥作用,调节胆汁酸、葡萄糖和磷酸盐的全身代谢。FGF通过与FGF受体(FGFRs)结合,激活丝裂原活化蛋白激酶(MAPK)和磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/AKT)等下游信号通路,从而影响各种细胞过程。
